Teucrium species possess perennial plants that are rich source of phenolic compounds. These compounds are employed to treat digestive and respiratory disorders and as diuretic, anti-septic, anti-rheumatic, anti-inflammatory, anti-oxidative, anti-microbial, anti-diabetic, hepatoprotective, hypolipidemic as well as anti-cancer agents1.
Teucrium have 11 species in Libiya and among them, 5 are endemic and problems are found at different taxonomic levels in these species. Moreover, this plant is represented in Libya through three sections and Polium is considered as the most challenging and heterogeneous section2.
Huge amount of human and environmental pressures is prevailing in Libya which cause genetic promotes genetic drift, leading towards more loss of genetic diversity. Hence, it’s the need of hour to evaluate the genetic diversity of these populations regarding both endemic as well as polymorphic species, as these evaluations were found to be vital to plan effectual conservation strategies3.
It is reported that both RAPD and ISSR are significant tools for these purposes. The RAPD is reasonable and yield results within short duration but it carry few drawbacks as well including low reproducibility4. Therefore, combining this technique with ISSR was found effective to distinguish among species because of its elevated reproducibility5.
Therefore, a new research was performed to evaluate the taxonomic associations and genetic diversity among Libyan Teucrium species based on both RAPD and ISSR6.
For this purpose, the genetic diversity was determined for 12 Teucrium taxa by RAPD and ISSR. Both the total and specific bands, polymorphism percentage, Shannon index (H) and Simpson index (D) were evaluated and the data was processed with cluster as well as principal component analyses.
This investigation revealed that T. capitatum and T. polium are two separate species. Moreover, it was found that Teucrium possess relatively low genetic diversity and approximately 42% of its species were endemic that need conservation priority. Also, this study confirms the validity of both RAPD and ISSR in order to determine taxonomic relation and genetic diversity.
